Also known as: Anti-Obesity Drug 9604, hGH Fragment 176-191

AOD-9604 is a synthetic peptide fragment of amino acids 176–191 of human growth hormone (hGH). Specifically designed to mimic the lipolytic effects of hGH without its effects on tissue growth or glycemic control. Developed by Monash University (Australia).

Mechanism of action

  • Stimulates lipolysis (fat breakdown) in adipocytes via the beta-3 adrenergic pathway
  • Inhibits lipogenesis (new fat formation)
  • Does not increase IGF-1 or serum glucose (unlike full hGH)
  • Does not generate insulin resistance with prolonged use

Uses & indications

  • Body fat loss (especially visceral and abdominal)
  • Obesity as adjunct therapy
  • Osteoarthritis: Under investigation for cartilage regeneration

Effects

Positive effects: In Phase IIa human trials, modest total body fat reduction (~1.2 kg vs. placebo over 24 weeks at 1 mg/day). Favorable metabolic profile.

Adverse effects: Well tolerated in clinical trials. Mild headache, local injection site reactions.

Considerations & contraindications

  • Not approved by FDA/EMA for obesity indication
  • Phase III clinical trials were not completed
  • Classified as OTC drug in Australia (TGA, 2014) for some uses

Research peptide. Approved in Australia by TGA (2014) as OTC drug in certain formulations. Phase IIa completed; Phase III trials not concluded.
