Kisspeptin-10

Also known as: Metastin (fragmento), KP-10

Kisspeptin-10 (KP-10) is a decapeptide (10 amino acids) and is the most potent minimal active fragment of the KiSS1 gene. Kisspeptins are the master regulators or natural "switches" of the onset of puberty and adult reproductive function. When administered, it strongly stimulates the hypothalamic-pituitary-gonadal (HPG) axis, causing a robust, pulsatile release of GnRH, LH, and FSH.

Mechanism of action

  • Binds to the GPR54 receptor (kisspeptin receptor) in the hypothalamus.
  • Induces massive and immediate secretion of Gonadotropin-Releasing Hormone (GnRH).
  • This triggers the pituitary to secrete LH and FSH, which in turn signal the testes or ovaries to produce natural testosterone or estradiol respectively.
  • Also has metastasis-suppressing roles (hence its original name "Metastin").

Uses & indications

  • Replacement/Recovery Therapy (TRT/PCT): Helps restart natural testosterone production in men after endocrine suppressor use. Alternative to HCG.
  • Infertility: Safe ovulation induction in women.
  • Psychological: Improves sexual desire and reproductive behavior directly mediated by the brain.

Effects

Rapid and pronounced elevation of endogenous testosterone, increased sexual desire, temporary reversal of hypogonadism.

Considerations & contraindications

  • Has an incredibly short half-life (minutes); injection elevates hormones strongly but briefly.
  • Continuous or incorrect dosing can desensitize the GPR54 receptor, achieving the opposite effect (chemical castration). Its use must be careful and occasional.

Regulatory approval

Extensive research in endocrinology, but no global commercial drug yet.

Dosage

Due to its pulsatile profile, small doses of 100-300mcg are used intermittently (e.g., twice a week) to avoid receptor desensitization.
