TB-500

Also known as: Timosina Beta-4 sintética, Thymosin Beta-4

TB-500 is a synthetic peptide replicating the active region of Thymosin Beta-4 (Tβ4), a 43-amino acid protein present in virtually all human tissues. Predominantly synthesized in the thymus, Tβ4 plays key roles in actin cytoskeleton organization, cell migration, and tissue healing processes.

Mechanism of action

Mechanism of action:

  • Actin-G binding: Sequesters actin-G monomers, regulating cytoskeleton polymerization and cell motility
  • Cell migration promotion: Activates macrophages, keratinocytes, and fibroblasts for tissue repair
  • Angiogenesis: Stimulates new blood vessel formation (upregulates VEGF)
  • Stem cell differentiation: Promotes cardiac and neuronal progenitor differentiation
  • Anti-inflammatory properties: Inhibits neutrophil migration and reduces chronic inflammation

Uses & indications

  • Muscle injury recovery: Tears, contusions, chronic muscle fatigue
  • Tendon and ligament injuries: Patellar tendinopathy, Achilles tendinitis, sprains
  • Reduction of scar tissue post-surgical or traumatic
  • Cardioprotection: In animal post-infarction models
  • Chronic wound healing

Effects

Beneficial effects (animal evidence): Wound healing acceleration, muscle and tendon regeneration, reduction of post-surgical adhesions, potential cardioprotective effects.

Reported adverse effects: Minimal. Possible transient lethargy post-injection; in rats, very high doses showed no significant toxicity. Limited human data.

Considerations & contraindications

  • Not approved for human therapeutic use (FDA/EMA)
  • WADA: endogenous Tβ4 is not prohibited, but synthetic analogues may be on the watch list
  • Do not use during pregnancy or breastfeeding

Regulatory approval

Research peptide. Not approved for clinical use. Natural Tβ4 has been evaluated in Phase I/II trials for wound healing and cardiac repair (RegeneRx Biopharmaceuticals).

Dosage

Loading protocol: 2–2.5 mg twice/week × 4–6 weeks. Maintenance: 2–2.5 mg/week. Combined protocols with BPC-157 are common.

Lyophilized: −20°C. Reconstituted in bacteriostatic water: 4°C, use within 4 weeks.
